dc.description.abstractCutaneous melanoma is a malignant neoplasm originating from melanocytes and represents the leading cause of death among skin cancers due to its high metastatic potential and the resistance of tumor cells to conventional therapies, including chemotherapy, radiotherapy, and immunotherapy with immune checkpoint inhibitors. Plant-derived phenolic compounds have been investigated as promising adjuvants in this context, and ellagic acid (EA) is a natural polyphenol that stands out for its antioxidant, anti-inflammatory, antiproliferative, and pro- apoptotic profile documented across different tumor models. The present work aimed to synthesize the available experimental evidence on the antiproliferative and antineoplastic effects of EA on cutaneous melanoma, with emphasis on the molecular mechanisms identified in preclinical models and on the modulation of the purinergic system as an emerging perspective. Chapter 1 constituted a narrative review of the theoretical background, addressing the epidemiological, histopathological, and molecular aspects of melanoma, the limitations of conventional therapies, and the chemical structure, sources, bioavailability, and general antineoplastic mechanisms of EA across different tumor models, including its inhibitory action on the ectonucleotidases CD39, CD73, and NDPK-B. Chapter 2 configured an integrative literature review, with searches conducted in the PubMed/MEDLINE and SciELO databases, focusing on original experimental studies published between 2015 and 2025 that assessed the effects of EA on cutaneous melanoma models. After screening according to the eligibility criteria and the PRISMA 2020 flowchart, three studies composed the final sample. The findings indicate that EA reduces the viability of murine and human melanoma cell lines (B16, B16F10, A375, and WM115) in a dose- and time-dependent manner, induces G1 cell cycle arrest, promotes apoptosis, and inhibits cell migration and invasion. The molecular mechanisms identified encompass the dual allosteric modulation of PTEN with prolonged inactivation of the PI3K/AKT pathway and accumulation of p53/p21, inhibition of EGFR with reversal of epithelial-mesenchymal transition, and reversible mixed-type inhibition of tyrosinase. Synergism with cisplatin was documented in B16F10 cells, and the clinical relevance of EGFR inhibition was supported by a prognostic correlate in melanoma patients. The data converge toward the recognition of EA as a promising compound for preclinical and translational investigations in the context of cutaneous melanoma, with potential for use in future combined therapeutic strategies.pt_BR
dc.description.resumoO melanoma cutâneo é uma neoplasia maligna originada nos melanócitos e representa a principal causa de morte entre os cânceres de pele, em razão de seu elevado potencial metastático e da resistência das células tumorais às terapias convencionais, incluindo quimioterapia, radioterapia e imunoterapia com inibidores de checkpoint imunológico. Compostos fenólicos de origem vegetal têm sido investigados como adjuvantes promissores nesse contexto, sendo o ácido elágico (AE) um polifenol natural que se destaca pelo perfil antioxidante, anti-inflamatório, antiproliferativo e pró-apoptótico documentado em diferentes modelos tumorais. O presente trabalho teve como objetivo sintetizar as evidências experimentais disponíveis sobre os efeitos antiproliferativos e antineoplásicos do AE no melanoma cutâneo, com ênfase nos mecanismos moleculares identificados em modelos pré- clínicos e na modulação do sistema purinérgico como perspectiva emergente. O Capítulo 1 constituiu uma revisão narrativa de fundamentação teórica, abordando os aspectos epidemiológicos, histopatológicos e moleculares do melanoma, as limitações das terapias convencionais, a estrutura química, as fontes, a biodisponibilidade e os mecanismos antineoplásicos gerais do AE em diferentes modelos tumorais, incluindo sua ação inibitória sobre as ectonucleotidases CD39, CD73 e NDPK-B. O Capítulo 2 configurou-se em uma revisão integrativa da literatura, com busca realizada nas bases PubMed/MEDLINE e SciELO, com foco em estudos experimentais originais publicados entre 2015 e 2025 que avaliassem os efeitos do AE em modelos de melanoma cutâneo. Após triagem conforme os critérios de elegibilidade e fluxograma PRISMA 2020, três estudos compuseram a amostra final. Os achados indicam que o AE reduz a viabilidade de linhagens murinas e humanas de melanoma (B16, B16F10, A375 e WM115) de forma dose e tempo-dependente, induz parada do ciclo celular em G1, promove apoptose e inibe migração e invasão celular. Os mecanismos moleculares identificados abrangem a modulação alostérica dual do PTEN com inativação prolongada da via PI3K/AKT e acúmulo de p53/p21, a inibição do EGFR com reversão da transição epitélio-mesenquimal e a inibição reversível de tipo misto da tirosinase. O sinergismo com cisplatina foi documentado em células B16F10, e a relevância clínica da inibição do EGFR foi sustentada por correlato prognóstico em pacientes com melanoma. Os dados convergem para o reconhecimento do AE como composto promissor para investigações pré-clínicas e translacionais no contexto do melanoma cutâneo, com potencial de atuação em terapias combinadas futuras.pt_BR
