Resume: | Ocimum basilicum L. (Lamiaceae) o manjericão, é uma planta aromática com grande importância, usada em diversos segmentos devido seu óleo essencial, teor de linalol e antocianina. Na Biotecnologia, a cultura de células e tecidos vegetais utilizado na micropropagação, é importante para o melhoramento genético dessa espécie. A pesquisa objetivou o estabelecimento de um protocolo de micropropagação in vitro para o manjericão-roxo, as sementes foram imersas em pré tratamento de álcool 70% durante 3 minutos, após foram utilizadas as concentrações de hipoclorito de sódio de 1%, 2%, 4% e 6% durante 20 minutos, e água destilada para tratamento controle, para determinação da influência na taxa de germinação e contaminação. A introdução in vitro foi realizada com tratamento de 6% de hipoclorito de sódio. Para a análise estatística foi utilizado o teste de Tukey ao nível de 5% de probabilidade. Os valores de contaminação não foram significativos, pois sua ocorrência foi restrita ao tratamento controle (66,6%). Para a variável de sementes germinadas o teste de comparação de médias demonstrou que a maior taxa ocorreu no tratamento controle (89,67%), porém devido a contaminação este tratamento não é indicado. O melhor tratamento para a desinfecção das sementes foi a concentração de 6% de hipoclorito de sódio, onde 28,17% das sementes germinaram após 28 dias, as demais concentrações não apresentaram diferença estatística entre si, apresentando a maior média de 5,67%. Na introdução in vitro a taxa de germinação foi de 65,71% sem contaminações, com altura média de 4,3 cm (porção aérea) e 2 cm (raiz). |
Abstract: | Ocimum basilicum L. (Lamiaceae) the basil, is an aromatic plant with great importance, used in several segments due to its essential oil, linalool and anthocyanin content. In biotechnology, the culture of cells and plant tissue used in micropropagation is important for the genetic improvement of this species. The research aimed to establish an in vitro micropropagation protocol for purple basil, the seeds were immersed in 70% alcohol pre-treatment for 3 minutes, then sodium hypochlorite concentrations of 1%, 2%, 4% and 6% were used for 20 minutes, and distilled water for control treatment, to determine the influence on germination rate and contamination. In vitro introduction was performed with 6% sodium hypochlorite treatment. For statistical analysis, Tukey's test was used at 5% probability level. The contamination values were not significant, since itsoccurrence was restricted to the control treatment (66.6%). For the germinated seeds variable the mean comparison test showed that the highest rate occurred in the control treatment (89.67%), but due to contamination this treatment is not indicated. The best treatment for the disinfection of the seeds was with a concentration of 6% of sodium hypochlorite, where 28.17% of the seeds germinated after 28 days, the other concentrations showed no statistical difference between them, presenting the highest average of 5.67%. In the in vitro introduction the germination rate was 65.71% without contamination, with an average height of 4.3 cm (aerial portion) and 2 cm (root). |
