| Resume: | É amplamente aceita a ideia de que o estresse é um fator importante no desencadeamento de transtornos mentais, como a ansiedade. O estresse crônico é um potente indutor de dano oxidativo no sistema nervoso central, sendo a peroxidação lipídica um dos marcadores biológicos mais deletérios dessa condição. Este estudo investigou o efeito protetor da N-acetilcisteína (NAC), com foco exclusivo na integridade das membranas lipídicas encefálicas de peixes-zebra submetidos ao estresse crônico imprevisível (ECI) por 7 dias. Os animais foram submetidos ao protocolo de ECI durante 7 dias aos seguintes agentes estressores em horários e dias alternados: perseguição por rede, aquecimento ou resfriamento da água, aglomeração, três trocas consecutivas de tanque e exposição do dorso. No 7º dia, além da exposição ao estresse, os peixes foram submetidos ao tratamento ou não por imersão em N-acetilcisteína (NAC), fluoxetina (FLU) ou cetamina (KET) durante 20 minutos. 24 horas após o tratamento, os animais foram anestesiados e eutanasiados por resfriamento rápido, e o encéfalo foi coletado, preparado e armazenado para análise bioquímica. O dano oxidativo foi quantificado por meio dos níveis de substâncias reativas ao ácido tiobarbitúrico (TBARS). Nossos resultados demonstraram que o ECI promoveu um aumento robusto e significativo nos níveis de TBARS, indicando uma degradação severa dos lipídios de membrana no encéfalo. O tratamento com NAC, fluoxetina (FLU) ou cetamina (KET) não induziu dano lipídico em animais não estressados, entretanto, os tratamentos com FLU e KET não foram capazes de reverter a peroxidação lipídica em animais estressados. Apesar disso, o tratamento com NAC reverteu de forma sustentada o dano lipídico causado pelo ECI. Estes dados evidenciam que a NAC pode atuar de forma sustentada após uma exposição farmacológica, atuando como um agente neuroprotetor em transtornos mentais relacionados ao estresse, impedindo especificamente a lipoperoxidação decorrente do estresse prolongado. |
| Abstract: | The idea that stress is a major factor in the onset of mental disorders, such as anxiety, is widely accepted. Chronic stress is a potent inducer of oxidative damage in the central nervous system, with lipid peroxidation representing one of the most deleterious biological markers of this condition. This study investigated the protective effect of N-acetylcysteine (NAC) on the integrity of brain lipid membranes in zebrafish subjected to chronic unpredictable stress (CUS) for 7 days. Animals were exposed to the CUS protocol for 7 days using the following stressors applied on alternating days and at varying times: net chasing, water heating or cooling, crowding, three consecutive tank changes, and dorsal body exposure. On day 7, in addition to stress exposure, fish were either treated or not treated by immersion in N-acetylcysteine (NAC), fluoxetine (FLU), or ketamine (KET) for 20 minutes. Twenty-four hours after treatment, the animals were anesthetized and euthanized by rapid cooling, and the brain tissue was collected, processed, and stored for biochemical analysis. Oxidative damage was quantified by measuring thiobarbituric acid reactive substances (TBARS) levels. Our results demonstrated that CUS induced a robust, significant increase in TBARS levels, indicating severe degradation of brain membrane lipids. Treatment with NAC, FLU, or KET did not induce lipid damage in non-stressed animals; however, FLU and KET treatments were unable to reverse lipid peroxidation in stressed animals. In contrast, NAC treatment sustainably reversed the lipid damage caused by CUS. These findings demonstrate that NAC may exert sustained effects following a single pharmacological exposure, acting as a neuroprotective agent in stress-related mental disorders by specifically preventing lipid peroxidation resulting from prolonged stress. |
